In doing some research we are thinking of taking the alilaguna from Hotel Hesperia to the airport. It seems pretty straight forward to me. Since we have never been to Venice just wanted a little input.

Where's the closest alilaguna stop from your hotel? How far is it to walk from your hotel to the stop?
Less than a quarter of a mile.
The regular water buses (vaporettos?) may be faster but the Aliguna is probably more scenic for a great overview of Venice.

It is easy to get luggage on and off the vaporettos.
But the Alilaguna is the airport shuttle -- a couple of stops in town and then straight to the airport. I'll eat my hat if it is takes more time than the vaporetto (which, like any good bus service, keeps making stops).
Alilaguna takes 1:20 from San Marco. The Blue bus from Piazzale Roma takes :20. The obvious variable is your time to the bus by vaporetto and that depends on location and which line you take.

If that distance involves crossing canals/bridges, make sure you're capable of carrying your luggage up/down steps on the bridges.
The Alilaguna does take quite a long time to the airport. And from the dock, it's a good 5-min walk to the airport bldg.
Leave yourself plenty of time before your flight. My last experience (2.5 yrs ago) at Marco Polo airport was not great. Very long lines and very chaotic at check-in.
Hotel Hesperia is on Canale di Cannaregio quite near the Alilaguna Guglie stop. The schedule shows the trip from there would be 48 minutes to the airport.
The other best option would be a vaporetto from Guglie, going two stops to Piazzale Roma, and then a land bus. By the time you made the connections between the different modes of transport, it would probably take the same amount of time as Alilaguna.
No vaporetto line goes all the way to the airport. Luggage is allowed on vaporetti.

the best way to find out how to get to your hotel is to ask the hotel - they are used to this question.
that said, if your hotel hesperia is the one in cannaregio that I found on google, IMO it is very unlikely that they would recommmend the alilaguna as there are no adjacent alilaguna stops.
your best options are
the ACTV bus [covered by the normal vaporetto pass or a blue or orange venicecard] from the airport to Piazzale Rome, then no 1 vaporetto to Ferovia [one stop] walk along the lista da spagna to the street where your hotel is
the ATVO bus [not covered by any passes, E3 each, takes 20 rather than 30 minutes and luggage space under the bus, not in the bus itself] to PR, then vaporetto as above.
Water taxi from the aiport to your hotel - probably around E90.
or a water-taxi from the PR to your hotel - probably about 20E.
on our recent trip, as we were arriving quite late at night [which turned out to be even later due to flight delays] we got a water-taxi to our accommodation, and the vaporetto and bus back to the airport when we left.

ann--I was just at the Alilaguna site and they claim to have a Guglie stop on Linea Arancio.

yep they do, but the times are very restricted [we wanted to get the arancia from rialto when we left, but it stops at 16.30, far too early for our 8.30pm flight] and it's the "wrong" side of the canal for kmasden. but if the times are right, it is probably the best option.
i missed it as I didn't recongnise "guili" - I've only been past it once, and never used it.
actually the OP's best bet would be to arrive by train - then he could walk straight up the lista da spagna! shame there's no train from the airport.

Or, if the new Calatrava bridge is ready by then, walk right over to Piazzale Roma with little difficulty.

we didn't see much sign of it when we were there last month.
but carrying/dragging suitcases anywhere round Venice is no fun - it is definitely a place for travelling light.

annhig, we aren't in need of getting to the hotel from the airport that part is covered. We need to get to the airport form the Hotel. According to my search the Guglie stop is very close to Hotel Hespera. I was at the alilaguna website from what I can tell it seems the easiest. Just wanted to confirm.

That is the same info I came up with and it seems the boat leaves every hour in the afternoon.
Ellenem has given the correct information. Alilaguna Arancio Line boat from Guglie stop near Hespeia to Airport via Murano (this is relatively a new line, maybe since a few years? It does not go via San Marco at all so it's confusing that the Alilaguna's time table says Linea Arancio SAN MARCO - Aeroporto, as if San Marco = Venice). It has a limited timetable, 1st dep. Guglie at 08:16, runs every hour only till 14:16. Arrives at Airport dock in 48 minutes. If this time table is convenient to you, it is much easier (and quicker I suspect)than vaporetto to Piazzale Roma + land bus to airport.
