Eastern Canada

We are quite undecided. It is easier to plan trips to Europe and several cities. This is the last query. You may go directly to one of the major cities that are on the route between Toronto and Quebec and from there make all excursions? Would that be more comfortable? And what would that city? They could also be required two cities, say two days each and then do the tours. That would not be problem. But which? And travel between cities is more convenient by bus or train. We have to get tickets in advance? We are living from Boston or New York.
Again..thanks a lot.
