What is the best/fastest/easiest route to drive from Toronto to St. Adrew's-by-the-sea? I have no problem cutting through the US if necessary. How many "driving hours" can I expect the trip to take?

Wow, that's a long trip.
The mostly-freeway route through the NYThruway/Masspike/I-95 is about 1,500km.
Trans-Canada, which is mostly freeway until you leave the St. Lawrence River, is about 1,450km.
If you cut through Vermont & New Hampshire via I-93/NH112/US301, it's about 1,380km; but parts of it is quite slow.
In short, there's no easy or fast way. It's 15-16 hours of drive, by my estimate. Fine for a 2-day each direction.
