What is the best route and how easy is it?

Someone just asked about this a couple of days ago. rkkwan gave a very detailed answer about taking the ferry from the airport to Macau upon arrival in Hong Kong. If you do a search, you'll find it.
If you're connecting directly from a flight into HKG, you can use the direct HKG-Macau Turbojet service without going through HK immigration:

Otherwise, you can go to the pier on Hong Kong Island and take the regular and much more frequent Turbojet to Macau, or Cotaijet to Taipa (say if you're going to the Venetian). You can take the Cityflyer airport bus direct from airport to the ferry pier. HK$40, every 20 minutes, takes about 35 minutes.
Super Class has complimentary drinks and snacks; and has smaller, more exclusive, cabins. But the seats are similar if not identical.
