I was looking at booking the bus from HCMC to SR by the Mekong Express. It looks like we can make it in one day. Has anyone done this and do you book thru your hotel? Thanks for your help!!

It takes about 6 hours. Booking through your hotel should be ok but not all hotels will do this and some will charge. More up to date info on these sights:

You can book through your hotel or an agent. I would travel during the day rather than at night. I think you are looking at more like 10+ hours, as PP to SR is 5.
It takes 6 hours from Phnom Penh to Siem Reap.(Did that 12 months ago)
Agree with KimJapan that you would be looking at 10+ hours.
Avoid any night travel due to condition of the roads, and what may wander across them at night.
Thanks gearsau, did you depart from HCMC? There is a bus that departs HCMC at 7:30 arrives at PP at 13:30 and I read there are busses departing up to 14:00 for SR. Should I assume they coordinate these for travellers going this route? What time does it get dark, is this too late to leave, for travelling to Siem Reap? Thanks!
If you have time, why not stay at least one night in PP to tour the city? Our biggest regret when we went to Siem Reap was not having the time to see PP as well. The Cambodians are lovely people and we hope to get back to more thoroughly see the country.

Actually, I did the trip from Siem Reap to Phnom Penh, so, in effect, the reverse. Left Siem Reap at 8:30AM, and was in Phnom Penh at 2:30PM. That included 30 minute lunch stop. $12.00 on Paramount Express bus. They even played the movie " Killing Fields" on the way as well.
Have a day in Phnom Penh to see Killing Fields memorial, S-21 Prison, National Museum. I had three days and it was one day too much. I much prefer Siem Reap,and am going back for the 5th time in September. Never go in the dry season, as its far too dusty and the countryside looks awful.
July to September will be good as its all green and clean. Even the temples look better as the dust is washed off them every day.
