If I book a flight on American Airlines Vacation sight, can I then call them and get an upgrade to first class(really just interested in return only being first class) using mileage.
Or is there some way to do this when I actually book online?
If I am reading it correctly, I can upgrade using 15,000 points per person and $175 each.

AA flies a 767 from HNL - DFW. If it's the 763-300, biz is the highest class. If it's the 762-200, there is a first class, so, in this case, no, it's not the same.
You might want to book your vacation separately from the flight.
AA is good about choosing FF miles on a to/from basis. If you want coach one way, and first the next, it can be done as each segment is a separate, but connected flight.

No business class on Hawaii runs, just coach and first. (AA's 763s have two cabins - economy and business on overseas flights, economy and first on domestics. Their 767-200s used in premium transcons, e.g. LAX-JFK, have three cabins.)

I'm 99%+ sure that AA Vacations uses ordinary/upgrade-eligible fare classes for the air portion of the packages, but you might want to phone AAdvantage Customer Service and confirm that with them.
